    A Field Theoretical Approach to Stationarity in Reaction-Diffusion Processes

    The aim of the project is to devise a general method to characterise the stationary state in finite systems, subjected to particular microscopic, local dynamics. Specifically, we have investigated, by means of field-theoretic techniques, the non-universal properties of a single species reaction-diffusion system. To make the stochastic process accessible to field-theoretic methods we have taken the Doi-Peliti formalism, which provides an exact description of the process in terms of a field theory. Furthermore, alongside the analytical approach we have investigated the system by means of Monte Carlo simulations in C. Field-theoretical techniques, usually used for the computation of universal quantities at criticality, have been employed for the characterisation of non-universal properties of the stationary state. A general definition of the stationary state in terms of a field theory has been given and a strong agreement between analytical results and Monte Carlo simulations has been found, near and away from the critical domain

    Organizational system thinking as a cognitive framework to meet climate targets

    System thinking is a crucial cognitive framework to enable individual pro-environmental behavioral changes. Indeed, a large body of literature has shown a significant and positive association between individuals’ system thinking capacities and perceptions of the threat posed by climate change. However, individual behavioral changes play a limited role in addressing climate change compared to large organizations involved in a significantly larger share of economic activities. Do organizations exhibit system thinking capacities? Here, we conjecture that system thinking is a cognitive framework observable at an aggregated group level and, therefore, organizations, not just individuals, can exhibit characteristic levels of system thinking. We conceptualize a definition of organizational system thinking and develop an empirical method to estimate it using a large body of textual data from business organizations. Then, we show that system thinking organizations are more likely to lower emissions and align them with the pathways required to meet the climate targets set by the Paris Agreement. Finally, we discussed the theoretical and policy implication of our study. Overall, our results suggest that system thinking is a relevant organization-level cognitive framework that can help organizations align their emissions with global climate targets

    Reorganization of interaction networks modulates the persistence of species in late successional stages

    Ecological interaction networks constantly reorganize as interspecific interactions change across successional stages and environmental gradients. This reorganization can also be associated with the extent to which species change their preference for types of niches available in their local sites. Despite the pervasiveness of these interaction changes, previous studies have revealed that network reorganizations have a minimal or insignificant effect on global descriptors of network architecture, such as connectance, modularity and nestedness. However, little is known about whether these reorganizations may have an effect on community dynamics and composition.To answer the question above, we study the multi-year dynamics and reorganization of plant–herbivore interaction networks across secondary successional stages of a tropical dry forest. We develop new quantitative tools based on a structural stability approach to estimate the potential impact of network reorganization on species persistence. Then, we investigate whether this impact can explain the likelihood of persistence of herbivore species in the observed communities.We find that resident (early-arriving) herbivore species increase their likelihood of persistence across time and successional stages. Importantly, we demonstrate that, in late successional stages, the reorganization of interactions among resident species has a strong inhibitory effect on the likelihood of persistence of colonizing (late-arriving) herbivores.These findings support earlier predictions suggesting that, in mature communities, changes of species interactions can act as community-control mechanisms (also known as priority effects). Furthermore, our results illustrate that the dynamics and composition of ecological communities cannot be fully understood without attention to their reorganization processes, despite the invariability of global network properties

    An evaluation of the potential of Sentinel 1 for improving flash flood predictions via soil moisture–data assimilation

    open8siThe assimilation of satellite-derived soil moisture estimates (soil moisture–data assimilation, SM–DA) into hydrological models has the potential to reduce the uncertainty of streamflow simulations. The improved capacity to moni- tor the closeness to saturation of small catchments, such as those characterizing the Mediterranean region, can be exploited to enhance flash flood predictions. When compared to other microwave sensors that have been exploited for SM– DA in recent years (e.g. the Advanced SCATterometer – AS- CAT), characterized by low spatial/high temporal resolution, the Sentinel 1 (S1) mission provides an excellent opportu- nity to monitor systematically soil moisture (SM) at high spatial resolution and moderate temporal resolution. The aim of this research was thus to evaluate the impact of S1-based SM–DA for enhancing flash flood predictions of a hydro- logical model (Continuum) that is currently exploited for civil protection applications in Italy. The analysis was car- ried out in a representative Mediterranean catchment prone to flash floods, located in north-western Italy, during the time period October 2014–February 2015. It provided some important findings: (i) revealing the potential provided by S1- based SM–DA for improving discharge predictions, espe- cially for higher flows; (ii) suggesting a more appropriate pre-processing technique to be applied to S1 data before the assimilation; and (iii) highlighting that even though high spa- tial resolution does provide an important contribution in a SM–DA system, the temporal resolution has the most crucial role. S1-derived SM maps are still a relatively new product and, to our knowledge, this is the first work published in an international journal dealing with their assimilation within a hydrological model to improve continuous streamflow simulations and flash flood predictions.     O gerenciamento integrado dos resíduos sólidos é de fundamental importância para a qualidade de vida. O objetivo deste trabalho é analisar a atual situação da cidade de Chapecó (SC) no tocante ao gerenciamento de resíduos sólidos urbanos (RSU) e identificar quais são os destinos dados a esses subprodutos descartados pela população urbana da cidade. E ainda, comparar os métodos modernos e sustentáveis da atualidade com o modelo empregado pelas empresas coletoras da cidade. Foi realizado um comparativo entre as oito premissas de Sachs (2007) com o plano de gerenciamento de resíduos sólidos no município de Chapecó (SC). A abordagem da pesquisa é qualitativa e quantitativa, e consiste em uma pesquisa descritiva, pois visa observar, registrar e analisar os fenômenos já expostos. Os resultados evidenciam que o município está em um processo evolutivo com o que contempla a Política Nacional de Resíduos Sólidos, pois possui vários instrumentos voltados à efetivação do gerenciamento integrado dos serviços de coleta, segregação e destinação adequada dos RSU, dentre deles o plano nacional, estadual e municipal de resíduos sólidos. Além disso, são apresentadas neste estudo novas alternativas que podem ser desenvolvidas na cidade a fim de melhorar a qualidade do destino, reuso do produto através de processos de transformação, como por exemplo, por meio da utilização da matéria prima como adubação para correção do solo e transformação do gás liberado na atmosfera em energia elétrica

    Combined inhibition of p97 and the proteasome causes lethal disruption of the secretory apparatus in multiple myeloma cells.

    Inhibition of the proteasome is a widely used strategy for treating multiple myeloma that takes advantage of the heavy secretory load that multiple myeloma cells (MMCs) have to deal with. Resistance of MMCs to proteasome inhibition has been linked to incomplete disruption of proteasomal endoplasmic-reticulum (ER)-associated degradation (ERAD) and activation of non-proteasomal protein degradation pathways. The ATPase p97 (VCP/Cdc48) has key roles in mediating both ERAD and non-proteasomal protein degradation and can be targeted pharmacologically by small molecule inhibition. In this study, we compared the effects of p97 inhibition with Eeyarestatin 1 and DBeQ on the secretory apparatus of MMCs with the effects induced by the proteasome inhibitor bortezomib, and the effects caused by combined inhibition of p97 and the proteasome. We found that p97 inhibition elicits cellular responses that are different from those induced by proteasome inhibition, and that the responses differ considerably between MMC lines. Moreover, we found that dual inhibition of both p97 and the proteasome terminally disrupts ER configuration and intracellular protein metabolism in MMCs. Dual inhibition of p97 and the proteasome induced high levels of apoptosis in all of the MMC lines that we analysed, including bortezomib-adapted AMO-1 cells, and was also effective in killing primary MMCs. Only minor toxicity was observed in untransformed and non-secretory cells. Our observations highlight non-redundant roles of p97 and the proteasome in maintaining secretory homeostasis in MMCs and provide a preclinical conceptual framework for dual targeting of p97 and the proteasome as a potential new therapeutic strategy in multiple myeloma

    Molecular characterization of HIV-1 subtype C gp-120 regions potentially involved in virus adaptive mechanisms

    The role of variable regions of HIV-1 gp120 in immune escape of HIV has been investigated. However, there is scant information on how conserved gp120 regions contribute to virus escaping. Here we have studied how molecular sequence characteristics of conserved C3, C4 and V3 regions of clade C HIV-1 gp120 that are involved in HIV entry and are target of the immune response, are modulated during the disease course. We found an increase of "shifting" putative N-glycosylation sites (PNGSs) in the α2 helix (in C3) and in C4 and an increase of sites under positive selection pressure in the α2 helix during the chronic stage of disease. These sites are close to CD4 and to co-receptor binding sites. We also found a negative correlation between electric charges of C3 and V4 during the late stage of disease counteracted by a positive correlation of electric charges of α2 helix and V5 during the same stage. These data allow us to hypothesize possible mechanisms of virus escape involving constant and variable regions of gp120. In particular, new mutations, including new PNGSs occurring near the CD4 and CCR5 binding sites could potentially affect receptor binding affinity and shield the virus from the immune response. © 2014 Cenci et al

    The NA62 experiment will begin taking data in 2015. Its primary purpose is a 10% measurement of the branching ratio of the ultrarare kaon decay K+π+ννˉK^+ \to \pi^+ \nu \bar{ \nu }, using the decay in flight of kaons in an unseparated beam with momentum 75 GeV/c.The detector and analysis technique are described here.Comment: 8 pages for proceedings of 50 Years of CP
